>Resumen: The dorsal raphe nucleus (DR) controls
>forebrain serotonin (5-HT), regulating stress
>and emotional states. Dysregulation of DR 5-HT
>cells has been implicated in the pathophysiology
>of mood disorders including depression and
>anxiety. DR neurons are regulated by excitatory
>and inhibitory drives mediated by glutamate and
>GABA axons arising from different brain
>regions. These multiple origins suggest a very
>complex synaptic neuropil within the
>DR. However, we do not know much about how
>these axons are organized or whether they could
>be involved in any ordered synaptic
>arrangements, and this is in part due to
>technical limitations. But now we can address
>these questions using a recently developed
>high-resolution immunofluorescence technique
>called Array Tomography. This technique allows
>the quantitative analysis of the synaptic
>neuropil by studying multiple synaptic antigens
>as well as their relationships to each other in
>the same volume of tissue. Our study revealed
>new synaptic features of DR circuits that may
>underlie to a direct GABAergic presynaptic
>regulation of glutamate transmission, supporting
>a permissive role of GABA on DR’s excitatory drive.
